GTA V is well under way for a release date in 2012. There is sufficient evidence that it will take place in Los Angeles and its surrounding areas. Expect it to be greatly enhanced from the benchmark, GTA IV.

LA seems highly possible.
They've already got a lot of the map done with LA Noire, just need to touch it up a bit to make it look more modern.
