If you have a previous save before you blew it up no but if not there is a high chance you might have to. My guess would be that they wouldn't put it there if it becomes unreachable.

yeah you'll have to restart, that's why i usually look up guides on missable trophies and stuff so i dont have to restart the whole game to get one thing

Even so m8 its not that bad, if all you have to focus on in ur second playthrough is bobbleheads, you can easily clear through it a couple of hours.

Play the vault the same and get good items etc. to start with.
From your first level up focus solely on upgrades 2 help slaughter!!! :evil: A fun, bad playthrough is really quick, just kill anything that moves and loot anything that doesnt.

BUT, there may be hope, i remember seeing a fallout save on this site that helps to get the karma trophies.
Maybe use those hope it helps
