For update a jailbreak iDevice you need to restore, so yes you will lose everything you need to put everything back

first make sure you back up your phone before you do an update. you should be able to keep your app store apps but if you want to keep the cydia apps to best to download "pkgbackup" this will backup everything jailbroken on your phone. if you decide to jb again remember that 4.3.5 is a tethered jb.

If you have cracked apps then the answer is no. Only if you jailbreak on 4.3.5 but it is thetered. Stick to 4.3.3 and wait until iOS5 is avaiable then update because the hackers said they already have the untheter for iOS5.
