Well formatting it would remove everything. However HDD0 tells me that it is the first drive and probably the one with windows on it. So you wouldn't be able to format it as long as it was running. But yes if you were to format it you would have to re-install windows provided that was the drive with windows on it.
CCleaner is a good program to remove cached files and temp files. If it is a hard drive you can also defrag the drive within windows. Right click on the drive in My Computer and go to properties. From there go to tools and click Defrag / Optimize (depends on what OS you are on the wording is different)
