I believe you do need the eSata station, as that is what your hard drive will go in but I'm not sure. I never used one.
[Disclaimer] Do not take my word as a solid answer, wait for more replies especially from E3 Flasher users!

Yes you need it for downgrading, but after you have downgraded to whatever OFW or CFW you want successfully, you can remove the E3 Flasher from the PS3 and put back your HDD in like it was before. Whether you remove it or not is your choise. But remember wait for further answers. Happy [Disclaimer] Do not take my word as a solid answer.

you dont need an esata drive the e3 flasher it self is powered by the nor (info from alchybear <3) it will just power up and say storage could not be found and then stay there and you can continue to back up your bios and so on i think the esata drive was introduced with the dual boot but now that they have stopped selling the clip for dual boot its just a nice extra to have

Save Point
you use it to put Your HDD in to downgrade and stuff but you can leave the E3 Flasher attached to your PS3 if you want and just have the esata station read your HDD i would recommend leaving it attached to your console just incase you Brick or want to downgrade again

Do a barrel roll!
esata is like a dualboot! same as a pc haves, you can switch from linux to windows for example. same with the ps3: you can switch from a ps3ita to rebug for example
