I've got Rebug 4.80 CEX.

Should I convert to DEX for online play?

Also do I need to use SEN enabler or similar before online play or is it built in to DEX?

And finally,can I safely play online from external hard drive?

Converting to DEX is purely optional although i do personally recommend it, it's literally no different than CEX as you can still use CCAPI (Once it's been updated to work on 4.80 CFW that is lol) if that's what you use, only things are you'll need DEX EBOOTS for your games and you'll have to pick the CEX option when connecting tools if you use CCAPI on DEX. You can use SEN on CEX or DEX as it's not built into either. If you have games on your External HDD yes you can play them just as safely as on the Internal HDD.
