Nope the trophies do not pop on load I am affraid I had to play a level in "Classic" mode first and got a score of 430k which popped all of these.
I will be testing more just to be sure but trying to get this game finished is a pain
I am going to check if you can play 1 level and die and see what happens. I will post back here soon with the info for you.

Just went into a "Classic" mode level and died at the beginning and I got 3 trophies.
Slaughterific (Kill 1000 enemies)
Average Scorer (Score 50,000 on each mode)
High Scorer (Score 125,000 on each mode)
Now that being said the save I used was the top one and there is still 200+ enemies to kill before Massacretastic would pop anyways.
As for the other trophies without testing properly I assume that you only have to do 1 little thing from each trophy at the moment.
Obviously when the final save is uploaded it will make thing a lot easier for this but at the moment I am a little bit off from that haha.
I hope this makes it a little easier to understand. I might have to redo the Read Me files in the saves now

Not a problem at all. I am aiming to have the full save set made within the next 3 days so hopefully I will be able to do some proper testing when it is done and post an accurate description of what needs to be done.
I am just being apart of NGU and sharing is caring after all
